Peterzano claimed to be a pupil of the renowned master Titian, the leading figure of the Venetian school, and he often signed his works as "titiani alumnus" to reflect this prestigious tutelage. Peterzano's debut in Milan was marked by the counterfaçade frescoes in San Maurizio al Monastero Maggiore, completed in 1573, where his works demonstrated a blend of Mannerist and Venetian influences. Among his significant projects was the series of frescoes in the presbytery of Garegnano Charterhouse, created between 1578 and 1582, which are considered some of his masterworks. Another notable work is the "Nativity with Saints and Angels" in the church of Santa Maria di Canepanova in Pavia.

One of Peterzano's most notable contributions to art history is his role as a teacher to the famed Caravaggio. In 1584, Peterzano signed a contract to take on a 13-year-old boy named Michelangelo Merisi as an apprentice. Merisi would later become known as Caravaggio, a key figure in the Baroque movement. The apprenticeship, which lasted for four years, was pivotal in shaping Caravaggio's early artistic development.
